Amphetamines are synthetic drugs having a strong stimulant effect on the central nervous system. The basic structural unit of amphetamines comprises the phenyl ethylamine nucleus. Some amphetamine derivatives, also manufactured as illicit drugs, are 3,4-methylenedioxymethamphetamine (MDMA), also known as ecstasy, 3,4-methylenedioxyamphetamine (MDA), and methamphetamine having different stimulant effects. Some antidepressants and slimming agents such as Lida (containing sibutramine as active ingredient) also contain the amine active constituents like amphetamines. Although various spectroscopic techniques such as fluorescence, IR-Raman, and more sophisticated chromatographic methods such as HPLC, LC, and GC coupled to MS detectors exist in literature for the assay of these amine compounds, simple, flexible, sensitive and low-cost spectrophotometric methods have not been devised. Thus, the purpose of this work is to develop ion-associative complexes of these cationic amines with anionic basic dyes such as bromocresol purple (BCP), methyl orange (MeO), and eriochrome black-T (erio-T) in acidic medium, and to extract these colored complexes into suitable organic solvents for the quantitative estimation of these amines. The conditions for optimal extraction of analyte were determined for each given reagent. The optimal pH of ion-associative complexes extraction (experimentally found between pH 3,0 and 3,6, depending on the analyte and reagent) was usually greater than the pKa of reagent dye and smaller than the pKa of amine (ammonium) or close to these values. The extracting solvent was either pure chloroform or a dilute solution of buthanol in chloroform.The proposed method for MDMA, fluoxetine, sibutramine was statistically validated using ecstasy, prozac, lida against standard HPLC equipped with a Hypersil C-18 column with (70 % acetonitrile-30 % triethylamine aqueous solution in pH 6 o-phosphoric acid) mobile phase.

Amfetaminler, merkezi sinir sistemi üzerinde çok güçlü uyarıcı etkisi olan sentetik ilaçlardır. Amfetaminlerin temel yapısını feniletilamin çekirdeği oluşturur. Yasadışı uyuşturucular olarak üretilmiş 3,4-methylenedioxymethamphetamine (MDMA), ecstasy olarak bilinir, 3,4-methylenedioxyamphetamine (MDA) ve metamfetamin gibi bazı amfetamin türevleri birbirinden farklı uyarıcı etkilere sahiptir. Bazı antidepresanlar ve etken madde olarak sibutramin içeren Lida gibi zayıflama ilaçları da amfetaminler gibi aktif amin bileşenleri içerirler. Literatürde bu amin bileşiklerinin analizi için flouresans, IR-Raman gibi çeşitli spektroskopik metotlar ile HPLC, LC, GC ve bunların MS dedektörleri ile birleştirilmiş sistemleri gibi karmaşık kromatografik metotlar var olmasına rağmen basit, kolay uygulanabilir, hassas ve düşük maliyetli bir spektrofotometrik yöntem bulunmamaktadır. Bu kapsamda bu tez çalışmasının amacı; bahsi geçen katyonik amin bileşiklerinin erio-T, BCP (bromocresol purple) ve metil oranj gibi anyonik boyalar ile asidik ortamda meydana getirdiği iyon-asosiyatif komplekslerin oluşumunun sağlanması ve bu renkli komplekslerin uygun organik çözücülere çekilerek amin yapılı uyarıcı maddelerin kantitatif tayinlerinin gerçekleştirilmesidir. Çalışılan her reaktif için analit ekstraksiyonunun en uygun yapıldığı koşullar sağlanmıştır. İyon-asosiyatif komplekslerinin en uygun ekstraksiyon pH'ı (deneysel çalışmalar sonucunda analit ve reaktiflere bağlı olarak bulunan pH 3,0-3,6) genellikle reaktiflerin pKa değerlerinden büyük, aminlerin (amonyum) pKa değerlerinden küçük veya bu değerlere yakındır. Ekstraksiyon işleminde kullanılan organik çözücü saf kloroform ya da kloroform-bütanol karışımıdır. Geliştirilen metot MDMA, fluoksetin, sibutramin için ecstasy, prozac ve lida gerçek örnekleri kullanılarak istatistiksel olarak standart bir HPLC metoduna (Hypersil C-18 kolon ve mobil faz olarak % 70 asetonitril, % 30 o-fosforik asit kullanılarak pH'sı 6'ya ayarlanmış % 1'lik trietilamin çözeltisi kullanılmıştır) karşı valide edilmiştir.
