Human sperm DNA fragmentation has been proposed as one of the basic elements of male infertility. That affects IVF / ICSI (in vitro fertilization / Intracytoplasmic sperm injection ) adversely. There are numerous studies that sperm nuclear DNA damages are showing an association between infertility and pregnancy with formation.In the processes with assisted reproductive techniques (ART) sperm cryopreservation (sperm freezing ) may applied in the cases where it is essentials. Cryopreservation both increased success in assisted reproductive cure and testis surgery, it made contribution to protect following fertility before chemotherapy and radiotherapy. Common principles of gamete and embryo cryopreservation are; freezing the material after stabilized with cryoprotectant, storing in liquid nitrogen at - 196 °C and while thawing changing physical media where gamete and embryo can protect their aliveness after removing cryoprotectants. The significance is determining of sperm cryopreservation frequently used in ART after these processes how much damage gives related to DNA fragmentation. In case of such studies less harmful techniques may be improved so that ART results can be improved positively.One hundred (n=100) persons who are with normospermic sperm parameters (between 20-40 ages) were tested. Sperm DNA damage were evaluated with Halosperm Technique. Fresh sample were investigated in liquid form, the remaining was kept frozen and after one month it was thawed, re-evaluated with Halo sperm technique and the results were comprised.

İnsan sperm DNA fragmantasyonu, erkek infertilitesi ile ilgili temel göstergelerden biri olarak öne sürülmüştür ve IVF/ ICSI (in vitro fertilization/ Intracytoplasmic sperm injection) sonuçlarını olumsuz yönde etkilemektedir. Sperm nüklear DNA hasarları ile infertilite ve gebelik oluşumu arasında korelasyon olduğunu gösteren çok sayıda çalışma bulunmaktadır.Yardımcı Üreme Teknikleri (YÜT) ile yapılan işlemlerde gerekli durumlarda sperm kriyoprezervasyonuna (dondurma) başvurulmaktadır. Kriyoprezervasyon hem üremeye yardımcı tedavilerdeki başarıyı arttırmış, hem de testis cerrahisi, kemoterapi ve radyoterapiden önce, gelecekteki fertilitenin korunmasına katkıda bulunmuştur. Gamet ve embriyo kriyoprezervasyonunun genel prensibi; materyalin kriyoprotektanlarla dengelendikten sonra soğutulması, sıvı nitrojen içinde -196°C'de depolanması ve çözülürken de kriyoprotektanların uzaklaştırılarak gamet ve embriyoların canlılıklarını koruyabilecekleri fizyolojik ortamlara geçirilmesidir. Tezin önemi YÜT'inde sıklıkla kullanılan sperm kriyoprezervasyon işlemlerinden sonra, DNA fragmantasyonu açışından sperme hasar verip vermediğini saptayabilmektir. Bu şekilde gelecek çalışmalarda sperme daha az zarar veren teknikler geliştirilebilir ve böylelikle YÜT sonuçları pozitif yönde geliştirilebilir.Normospermik sperm parametresine sahip toplam yüz (n=100) kişi (21-39 yaş aralığı) kişi teste dahil edildi. Sperm DNA hasarı Halosperm Tekniği ile değerlendirildi. Taze örnek likefiye olduktan sonra çalışılıp kalan kısım dondurularak saklandı ve bir ay sonra çözülüp tekrar Halosperm Tekniği kullanılarak değerlendirilip sonuçlar karşılaştırıldı.
