The effect of vortioxetine on the nociceptive system: an animal experimental studyObjective: Pain is a significant symptom that can cause a decrease in people's quality of life. There are many agents used in its treatment and antidepressants are one of the important ones. It is also common for people with pain to have psychiatric disorders. Many antidepressants have been shown to have analgesic activity. Vortioxetine is an antidepressant used in various psychiatric disorders, but its effect on the nociceptive system is unknown. The aim of this study was to investigate the effect of vortioxetine on nociceptive system.Method: A total of 40 mice were included in the study, and they were divided into four groups, each consisting of 10 mice. Group 1 was given distilled water whereas vortioxetine dissolved in distilled water was administered intraperitoneally to groups 2, 3 and 4 at a volume of 5 mg/kg, 10 mg/kg, and 20 mg/kg, respectively. Following the injection, mice were placed on the hot plate at 30 min and 90 min. The time taken to observe a hind paw licking or jumping was determined as the pain threshold. For mice that did not show hind paw licking or jumping response within 30 seconds, the experiment was terminated to prevent damage to the mice and the pain threshold was considered 30 seconds. Dunnett's test was used to evaluate the effect of time on the dose and one-way analysis of variance (ANOVA) was used to determine the effect of dose in 30 min and 90 min. Results: Post-hoc comparisons made by using the Dunnett's test showed that values at 30 minutes time interval were found to be higher in group 1 (24.85±1,138) than other groups (group 2: 19,4920±1,82316; group 3: 16,9210± 1,57698; and group 4: 12.25 ±1.33) (p<0.001). Similarly, values at 90 minutes time interval were found to be higher in group 1 (24.13±1.182) than other groups (group 2: 17.47±1.55; group 3: 14.33±1.78; and group 4: 9.65±1.51) (p0.001). Analysis of variance made between groups revealed that there was a significant difference between the doses at the 30th- and 90th-minute measurements (F[3, 36] = 109.93) (p<0.001).Conclusion: The findings obtained in this study have showed that vortioxetine given at doses of 5 mg, 10 mg, and 20 mg may decrease pain threshold and increase the sensitivity to pain. This sensitivity to pain may be due to the mechanisms of hyperalgesia or the cognitive effects of vortioxetine.
