The aim of the thesis is to develop an application which is providing the calculation of the probable suitable donor rates in Turkish society for a transplant patient who has Human Leukocyte Antigen (HLA) antibody positivity. In this application, Calculated PRA (CPRA) values are data for patients who have positive HLA antibody, with HLA tissue type of information belonging to 5 locus of 1192 healthy individuals without relationship with each other. The data used are obtained from Yıldırım Beyazıt University Dışkapı Training and Research Hospital Tissue Typing Laboratory. A patient waiting for organ transplant has anti-HLA antibody or not and how much of its positivity is determined by the method of Panel Reaktif Antikor (PRA). Ratio of antigen used in PRA panel varies from society to society. As there is no PRA panel specifically designed for our country, panels prepared for the European society, considered to be close to the Turkish community, are used. In this case, when the patient is put in the waiting list for organ, as it doesn't reflect the correct PRA positivity literally, it leads to misinterpretations in patient selection. In this study, allele and haplotype frequencies have been calculated for Turkish society and an application that makes CPRA value has been developed by using these frequency values. From the obtained data, allele and haplotype frequencies have been calculated with EM algorithm in arlecore which can be operated as a script version of Arlequin 3.5.1.3. Also using the obtained frequency values, a desktop and a web application which make CPRA calculation have been developed. Applications have been developed by using C# programming language in Microsoft Visual Studio 2010 programme. When this data has been analyzed, it has been realised that there are 19 HLA-A, 29 HLA-B, 13 HLA-C, 13 HLA-DRB1 and 5 HLA-DQ allelle types in our society. It has been also realised 270 different haplotypes for 5 locus haplotype. Allele frequencies obtained from the calculation results, have been compared with the frequency of the 3 European countries (Germany, Italy and France).

Veriler analiz edildiğinde, toplumumuzda 19 HLA-A, 29 HLA-B, 13 HLA-C, 13 HLA-DRB1 ve 5 HLA-DQ allel türünün bulunduğu görülmüştür. 5 lokus haplotip için ise 270 farklı haplotip görülmüştür. Hesaplama sonucunda elde edilen allel frekanslarının, 3 Avrupa ülkesinin (Almanya, İtalya ve Fransa) frekans değerleri ile karşılaştırması yapılmıştır.
