In the study, 28 Swiss Albino male mice were used to investigate theprotective effect of silymarin againts formaldehyde-induced oxidative testicular damage in mice. The animals were divided into four groups, control (C), formaldehyde (F), silymarin (S) andsilymarin+formaldehyde (SF), with equal numbers of animals in each group. In group C;intraperitoneal serum, in group F; 10 mg / kg / day formaldehyde, in group S; 100 mg/ kg / day silymarin and in group SF; 4 mg / kg formaldehyde +100 mg / kg / daysilymarin was given for 14 days. Plasma was separated from the blood samplesobtained at the end of the applications. The samples were stored at -20 °C in freezinguntil paraoxonase and arylesterase enzyme activities and HDL (high-densitylipoprotein) concentration were measured. Testicular tissue samples were sent to thepathology laboratory for histopathological examinations in 10% formaldehydesolution. In our study, a significant increase in both PON1 and ARE activity (P <0.05)was observed in the group given silymarin. This increase may be due to the fact thatsilymarin may be associated with increased antioxidant activity by synergistic actionwith PON1 and ARE enzymes. Similarly, in the formaldehyde treated group, asignificant decrease (P< 0.05) in the activity of PON1 and ARE could be suggested to berelated to the oxidative stress that may be caused by formaldehyde. In the group silymarin+formaldehyde, histopathological parameters were close to thecontrol and silymarin group. We believe that these positive effects of silymarin ontesticular cells may be related to the strengthening of the antioxidant defense systemin parallel with the changes in PON1 and ARE activities. As a result, it has been observed that intraperitoneal formaldehydeadministration in mice may cause cell damage and oxidative stress, whereas antioxidant silymarin causes protective effects in reducing testicular cell damage andpreventing oxidative stress. Keywords: Silymarin, Testicular damage, Paraoxonase, Arylesterase, Oksidatif stres.
