The present study was designed to determine in vitro whether Vaccinium myrtillus L. and Rubus idaeus L. fluid extracts have antimutagenic effects in human peripheral lymphocytes by means of chromosome aberration (KA), micronucleus (MN) and mitotic index (MI) tests. In the study, 10 groups were formed with peripheral blood samples taken from 6 healty individuals as a material. Fruit and leaf mixture extracts of V. myrtillus L. and Rubus idaeus L. were used as the test substance. Blood samples of the negative control group (Group I) were allowed to react in a cell culture medium without any treatment, while positive control group (Group II) was allowed to interact in a cell culture medium supplemented with mitomycin-C (MMC) at a dose of 0.3 μL /mL for each chromosome medium. Blood samples of the other groups were allowed to react for 24 hours with 0.3 μL /mL MMC and Vaccinium. myrtillus L. (Group III, IV, V and VI), and Rubus idaeus L. (VII, VIII, IX and X) extracts in 0.2 μL /mL, 0.4 μL /mL, 0.8 μL /mL and 1.6 μL /mL doses for each of cell cultures. At the end of the incubation period, culture cells were evaluated by chromosomal aberrations, mitotic, micronucleus and nuclear division index tests. Compared with Group II, it was determined that mitotic, nuclear division and nuclear cytotoxic cleavage indices were increased when chromosome aberrations and micronucleus indexes were decreased in extract groups. As a result, it was concluded that the fruit and leaf extracts of V. myrtillus L. and Rubus idaeus L. as test substances in the study had antimutagenic effects on the human peripheral lymphocyte cells at the working doses.

Bu çalışmanın amacı, Vaccinium myrtillus L. ve Rubus idaeus L. sıvı ekstraktlarının in- vitro olarak, insan periferal lenfositlerinde antimutajenik etkiye sahip olup olmadıklarını kromozom aberasyonu (KA), mikronukleus (MN) ve mitotik indeks (Mİ) testleri ile belirlemektir. Çalışmada test maddesi olarak V. myrtillus L. ve R. idaeus L.'nin meyve ve yaprak ekstraktları, materyal olarak da sağlıklı 6 bireyden alınan periferik kan örnekleri ile 10 grup oluşturuldu. Negatif kontrol grubu (Grup I) kan örnekleri hiçbir muamele yapılmayan hücre kültürü ortamında, pozitif kontrol grubu (Grup II) kanları ise her kromozom mediumu için 0.3 μg/ml dozunda mitomisin-C (MMC) ilave edilen hücre kültürü ortamında etkileşime bırakıldı. Diğer grupların kan örnekleri hücre kültürüne 0.3 μg MMC/ml ve 0.2 μL /mL, 0.4 μL /mL, 0.8 μL /mL ve 1.6 μL /mL'lik dozlarda V. myrtillus L. (Grup III, IV, V ve VI,) ve R. idaeus L. (VII, VIII, IX ve X) sıvı ekstraktları bulunacak şekilde 24 saat süre ile etkileşime bırakıldılar. İnkübasyon süresi bitiminde kültür hücreleri kromozom aberasyonları, mitotik, mikronükleus ve nükleer bölünme indeks testleri ile değerlendirildi. Grup II ile karşılaştırıldığında ekstrakt gruplarında kromozom aberasyonları ve mikronükleus indeksinin azaldığı saptanırken, mitotik, nükleer bölünme ve nükleer sitotoksik bölünme indekslerinin artmış olduğu belirlendi. Sonuç olarak, çalışmada test maddesi olarak V. myrtillus L. ve R. idaeus L.'nin meyve ve yaprak ekstraktlarının çalışılan dozlarda insan periferal lenfosit hücrelerinde antimutajenik etkilere sahip olduğu belirlendi.
