The aim of this study to evaulate the effects of two different force magnitudes on periodontium in rats. With the help of split mouth study design, one side was chosen randomly as experimental side and the other was control. Appliance was actived with 10 or 60 cN force magnitude and each group of rats were killed at day 1,7,21 and 42 to reflect all the phases of tooth movement. Distance between 1. molar and incisors was measured for experimental and control side at day 0,1,2,7,14,21,28,35 and 42 then actual orthodontic tooth movement was calculated. Data was analysed with Mann Whitney U test and there was no statistically significant difference between 10 and 60 cN groups for insicor-molar distances at experimental and control sides also for actual tooth movement at each time interval. Histological and immunhistochemical evaluation was made on upper 2. molar mesiobuccal root. Blocks were stain with hematoxylin eosin to measure PDL width in mm from both resorption and aposition sides with the help of soft imaging system. There was statistically significant difference only in aposition side at day 21 between 10 and 60 cN groups for PDL width. For immunhistochemical evalution, primary antibodies were used and resorption and aposition sides were analysed and counted positive cells stained by primary antibodyies. Data was examined with tukey post hoc tests and statistically significant differences were found for Bax primary antibody at day 1 in resorption side, Bcl2L1 primary antibody at day 7 in aposition side, RANK primary antibody at day 7 and 21 in resorption side and RANKL primary antibody at day 21 resorption side between 10 and 60 cN groups. As a result of this, we recommend that, minimal force magnitude that could cause tooth movement should be used as orthodontic force.Key Words: ortodontic tooth movement, orthodontic force light/heavy, rat, periodontium, immunhistochemistry.
