In this study, isolation of yeasts from traditional Turkish sucuk, their genotypical and phenotypical identification, determination of their functional and technological properties and selection of a yeast starter culture combination proper to characteristics of sucuk by using those yeasts in sucuk manufacture were aimed. For this purpose, 35 retailed fermented sucuk samples were collected from different regions of Turkey. Firstly, some microbiological (Total mesophilic aerobic bacteria (TMAB), total coliform, Micrococcaceae, lactic acid bacteria (LAB), yeast and mold) and chemical (pH and water activity (aw)) properties of the sucuks were determined. Afterwards, different yeast colonies were selected and isolated from the petri dishes on which the yeasts were counted; thereby, pure yeast isolates were obtained. Pure yeast isolates were identified phenotypically using mini API system with API ID 32C kits. Following the extraction of DNA from those isolates, kindredness and differences between the yeast species were determined using rep-PCR and RAPD PCR techniques. Sequence analysis was performed by multiplying the yeast isolates according to 26S rRNA regions and their DNA sequence was genotypically identified by scanning in the yeast library. Candida zeylanoides and Debaryomyces hansenii were found to be the most abundant species among the yeast isolates. Some strains (total 47 strains) were selected after isolation and their functional and technological properties were determined. In the final stage of the study, two strains which exhibit favorable functional and technological properties were selected from those two yeast species. Their different combinations were used in sucuk manufacture and effects on some characteristic properties of sucuk such as physicochemical and microbiological properties, volatile aroma components, textural and sensorial parameters were determined. Yeast counts varied between 2.15 and 7.89 log cfu/g in the retail sucuk samples. aw and pH values were found to be 0.71-0.93 and 4.69-6.58 in the samples, respectively. Total 255 yeast isolates were obtained from the traditional sucuk samples. As a result of the phenotypic identification, dominant yeast genus was determined to be Candida, while Cryptococcus, Geotrichum, Rhodotorula and Zygosaccharomyces were also detected. In the overall isolates, most abundant species determined phenotypically were C. zeylanoides (95 isolates), C. famata (its telemorph is D. hansenii) (75 isolates) and C. lipolytica (19 isolates). Verification of the identified yeast isolates were performed with the molecular techniques and Candida species were again the main yeast isolates. In addition to Candida, some yeast species belonging to Debaryomyces, Cryptococcus, Torulaspora, Clavispora, Yarrowia, Kluyveromyces, Pichia, Sporobolomyces, Rhodotorula, Kazachstania, Issatchenkia, Filobasidium and Wickerhamomyces genera were also detected. The identification revealed that C. zeylanoides and D. hansenii were the dominant two species. These two species were found to be resistant to 5%, 10% and 15% of salt and 0.1%, 0.3%, 0.5% and 1.0% of bile salt concentrations. Some strains of C. zeylanoides grew at 35 ºC and pH 2 while D. hansenii was unable to grow at pH 2, however some D. hansenii strains were able to grow at 35 ºC. Both strains were resistant to potassium sorbate and sodium benzoate in the concentrations of 0.1%, 0.3%, 0.5% and 1.0% while they were sensitive against natamycin and nystatin. All strains of those species exhibited lipolytic and proteolytic activity and catalase (+) properties. 3 and 4 strains of C. zeylanoides had ß-hemolysis activity and nitrate reduction ability to nitrite, respectively. D. hansenii did not possess ß-hemolysis, nitrate reduction and hydrogen sulfur production properties; however, all strains analyzed had gelatin hydrolyzing ability. Following the determination of functional and technological properties of those yeast species, C. zeylanoides S15c and D. hansenii S3c were selected for sucuk manufacture. Then 8 different sucuk samples were produced using the yeast species as single and combined starter cultures and/or LAB (Lactobacillus sakei, Lb. curvatus and Lb. plantarum). In first, 6th and 12th days of the ripening period, some microbiological, physical and chemical properties of the sucuk samples were determined. Yeast and LAB counts ranged from 4.78 to 7.01 log cfu/g and from 9.89 to 10.17 log cfu/g in the 12th day of the ripening, respectively. Moisture and aw levels of the samples decreased with the extending ripening period and those parameters were lower in the sucuks fermented with only yeast cultures; therefore, it was confirmed that proper drying took place in those samples. pH levels of the samples decreased rapidly in the 6th day of the ripening while slight increases occurred until the end of ripening. Generally, nitrite levels decreased during the ripening while TBA values dropped in the 6th day; however it increased in some samples at the end of the ripening. Color values of the sucuk samples were measured in 2 different points, internal part and outer surfaces. a* parameter indicating the redness of the samples ranged from 16.77 to 18.39 and from 11.74 to 15.80 in the internal part and outer surfaces, respectively. Most of the volatile compounds in the sucuk samples were sulphure containing components and terpenes. No ketones were obtained while limited numbers of aldehydes and ketones were observed. In the meanwhile, major volatile compounds were cuminaldehyde, di-2-propenyl disulfide, ?-terpinene and p-cymene. At the end of ripening, texture profile analysis (TPA) were carried out to determine the textural properties of the sucuk samples and their hardness, adhesiveness, gumminess and chewiness properties were found to be variable among the samples. In the case of sensorial evaluation, yeast fermented samples received higher scores compared to other samples. In conclusion, yeasts isolated from sucuks were identified in detail and functional and technological properties of the dominant yeast species were revealed. It was determined that the use of yeasts as starter culture in addition to LAB for ripening of sucuk could make significant contribution to the quality parameters of Turkish sucuk. Yeast starter culture enabled a better moisture and weight loss, decreased lipid oxidation and enhanced textural and color properties of the sucuks. Additionally, sucuk samples containing yeast cultures were found to be more acceptable organoleptically.

Bu araştırmada, geleneksel Türk sucuklarından mayaların izole edilmesi, bunların fenotipik ve genotipik olarak tanımlanması, fonksiyonel ve teknolojik özellikleri dikkate alınarak seçilen bazı mayaların sucuk üretiminde kullanılması ve sucuğun karakteristik özelliklerine uygun olanların starter kültür olarak belirlenmesi amaçlanmıştır. Bu amaçla Türkiye 'nin farklı illerinden geleneksel olarak üretilen 35 fermente sucuk örneği toplanmıştır. İlk olarak sucukların bazı mikrobiyolojik (Toplam mezofilik aerobik bakteri (TMAB), toplam koliform, Micrococcaceae, laktik asit bakterileri (LAB), maya ve küf) ve kimyasal (pH ve su aktivitesi (aw) özellikleri belirlenmiştir. İkinci aşamada maya sayımı yapılan petrilerden birbirinden farklı maya kolonileri seçilerek saflaştırılmış ve böylece saf maya izolatları elde edilmiştir. Elde edilen saf maya izolatları API ID 32C kitleri kullanılarak mini API sisteminde tanımlanmıştır. Daha sonra bu izolatlardan DNA ekstraksiyonu sonrasında, rep-PCR ve RAPD PCR teknikleri kullanılarak maya türleri arasındaki akrabalık ve farklılıklar belirlenmiştir. Farklılıkları belirlenen maya izolatlarının 26S rRNA bölgesi çoğaltılarak dizi analizi yapılmış ve elde edilen DNA dizisi kütüphanede taranarak izolatların genotipik olarak tanımlamaları yapılmıştır. Tanımlanan bu maya izolatlarından en fazla bulunan iki türün Candida zeylanoides ve Debaryomyces hansenii olduğu belirlenmiştir. Bu iki türün yüksek dilüsyonlardan saflaştırılan bazı suşları (toplam 47 suş) seçilerek bunların fonksiyonel ve teknolojik özellikleri belirlenmiştir. Çalışmanın son aşaması olarak seçilen bu iki türden fonksiyonel ve teknolojik özellikleri iyi olan iki adet suş belirlenmiştir. Bu iki suş farklı kombinasyonlarda sucuk üretiminde kullanılmış ve bunların sucuğun bazı fizikokimyasal ve mikrobiyolojik özellikleri ile uçucu aroma bileşenleri, tekstür ve duyusal özellikleri gibi bazı karakteristik özelliklerine etkisi saptanmıştır. Piyasadan temin edilen sucuk örneklerinin maya sayılarının 2.15-7.89 log kob/g arasında olduğu tespit edilmiştir. Örneklerin aw ve pH değerlerinin sırasıyla 0.712-0.932 ve 4.69-6.58 arasında olduğu belirlenmiştir. Geleneksel sucuk örneklerinden toplam 255 adet maya izolatı elde edilmiştir. Fenotipik tanımlama sonucunda hakim maya cinsinin Candida olduğu, bunun dışında Cryptococcus, Geotrichum, Rhodotorula ve Zygosaccharomyces cinsi mayaların da bulunduğu saptanmıştır. Toplam izolat sayısı içerisinde en fazla C. zeylanoides (95 izolat), C. famata (telemorfu D. hansenii) (75 izolat) ve C. lipolytica (19 izolat) 'nın olduğu fenotipik olarak tespit edilmiştir. Tanımlanan bu maya türlerinin doğrulanması moleküler tekniklerle genotipik olarak yapılmış ve yine majör maya türlerinin Candida türleri olduğu belirlenmiştir. Bunun dışında Debaryomyces, Cryptococcus, Torulaspora, Clavispora, Yarrowia, Kluyveromyces, Pichia, Sporobolomyces, Rhodotorula, Kazachstania, Issatchenkia, Filobasidium ve Wickerhamomyces gibi maya cinslerinin bazı türleri de saptanmıştır. Bu yöntemle tanımlama sonucunda C. zeylanoides ve D. hansenii 'nin hâkim iki tür olduğu tespit edilmiştir. Bu iki türün %5, 10 ve 15 tuz konsantrasyonlarına ve %0.1, 0.3, 0.5 ve 1.0 safra tuzuna karşı dayanıklı olduğu belirlenmiştir. C. zeylanoides türüne ait bazı suşların 35°C ve pH 2 'de bile geliştiği belirlenirken, D. hansenii 'nin pH 2 'de gelişemediği bununla birlikte bazı suşlarının 35°C 'de geliştiği saptanmıştır. Yine bu araştırmada her iki türün %0.1, 0.3, 0.5 ve 1.0 konsantrasyonlarındaki potasyum sorbat ve sodyum benzoata karşı dirençli, natamisin ve nistatine karşı ise hassas oldukları tespit edilmiştir. Yine iki türün analiz edilen tüm suşlarının lipolitik, proteolitik aktivite ve katalaz (+) özellik gösterdiği belirlenmiştir. C. zeylanoides 'in 3 suşunun ß-hemoliz aktivitesinin olduğu ve 4 adet suşunun ise nitratı nitrite indirgeme yeteneğinin bulunduğu saptanmıştır. D. hansenii 'nin ß-hemoliz, nitrat indirgeme ve hidrojen sülfür (H2S) üretme özelliğinin olmadığı, bununla birlikte analiz edilen tüm suşlarının jelatini hidroliz etme yeteneğine sahip olduğu tespit edilmiştir. Bu iki maya türünün fonksiyonel ve teknolojik özellikleri belirlendikten sonra C. zeylanoides S15c ve D. hansenii S3c suşları sucuk üretimi için seçilmiştir. Daha sonra bu maya türleri tek tek, kombine halde ve/veya LAB (Lactobacillus sakei, Lb. curvatus ve Lb. plantarum) 'lerin kullanıldığı 8 farklı sucuk üretimi gerçekleştirilmiştir. Bu sucukların olgunlaşmasının ilk, 6. ve 12. günlerinde örneklerin bazı mikrobiyolojik, fiziksel ve kimyasal özellikleri belirlenmiştir. Olgunlaşmanın 12. gününde maya ve LAB sayılarının sırasıyla 4.78-7.01 ve 9.89-10.17 log kob/g düzeylerinde olduğu belirlenmiştir. Nem ve aw değerlerinin olgunlaşma süresinin artmasıyla birlikte düştüğü tespit edilmiştir. LAB ilaveli sucuk örneklerinin nem ve aw değerlerinin daha düşük olduğu saptanmıştır. Örneklerin pH değerlerinde 6. gün sonunda hızlı bir düşüş ve 12. gün sonunda ise kısmi bir yükselmenin olduğu belirlenmiştir. Genel olarak nitrit değerleri olgunlaşma ile birlikte kısmi bir azalış gösterirken TBA değerleri 6. günde azalmış ancak 12. günde ise bazı örneklerde biraz artmıştır. Sucuk örneklerinin renk değerleri kesit yüzeyi ve dış yüzey olmak üzere 2 farklı şekilde ölçülmüştür. Bunun sonunda kırmızlığın göstergesi olan a* parametresi sucuk örneklerinde kesit yüzeyi ve dış yüzey için sırasıyla 16.77-18.39 ve 11.74-15.80 aralığında bulunmuştur. Sucuk örneklerinde uçucu aroma bileşenlerinin çoğunluğunun kükürtlü bileşenler ve terpenler olduğu saptanmıştır. Ketonlardan hiçbir bileşik bulunamazken aldehitlerin ve esterlerin ise az sayıda oldukları tespit edilmiştir. Sucuk örneklerinde en fazla bulunan uçucu bileşenlerin cuminaldehyde, di-2-propenyl disulfide, ?-terpinene ve p-cymene olduğu belirlenmiştir. Olgunlaşmanın son gününde sucuk örneklerinin tekstürel özelliklerini belirlemek için Tekstür Profil Analizi (TPA) yapılmış ve sonuçta sertlik, iç yapışkanlık, sakızımsılık ve çiğnenebilirlik özelliklerinin örnekler arasında değişkenlik gösterdiği tespit edilmiştir. Sucuk örnekleri duyusal olarak değerlendirildiğinde, maya ilaveli örneklerin daha fazla tercih edildiği saptanmıştır. Sonuçta farklı sucuklardan izole edilen mayaların detaylı tanımlanması yapılarak en fazla bulunan maya türlerinin fonksiyonel ve teknolojik özellikleri ortaya konmuştur. Sonuçta sucuk üretiminde LAB 'ne ek kültür olarak maya starter kültürünün kullanımının Türk tipi fermente sucuğun kalite özelliklerine önemli katkılarının olabileceği belirlenmiştir. Maya starter kültür kullanımı ile sucuğun nem ve ağırlık kaybı ile lipid oksidasyonunun azaldığı, tekstürel ve renk özelliklerinin iyileştiği tespit edilmiştir. Ayrıca maya starter kültür ilaveli pişmiş sucuk örneklerinin duyusal yönden daha çok beğenildiği gözlemlenmiştir.
