In this study, in the light of information obtained from previous researches, Lactobacillus acidophilus KPb4b strains, the potantial probiotic properties of which were identified in the culture collection of Food Microbiology Research Laboratories Department of Faculty of Engineering and Architecture of Bolu Abant İzzet Baysal University and confirmed by molecular genetic methods, were used. In the scope of this study, microencapsulation was carried out by the emulsion technique of potential strain, electrostatic vibration / dripping method and double layer coating (second layer coating with chitosan on the first coating performed by the electrostatic vibration / dripping method). At the end of the study, it was tried to determine the appropriate parameters to be used to protect the viability of microorganism by using the response surface methodology.According to the results of the study, the difference in the number of viable microorganisms at the beginning and at the end of the analysis was low, indicating that the resistance of the bacteria to the analysis conditions was high.According to the results of the study, the effect of aljinat concentration was very important (p<0.01), and the interaction effect of aljinat concentration and chitosan concentration) was also very important (p<0.01).According to the results of the study, it was concluded that the alginate concentration and calcium chloride molarity in the coating and double layer (electrostatic vibration/dripping method + kitosan) coating methods significantly influenced the microencapsul size as a consequence of (p<0.01) and besides it was detected that aljinat concentration was significantly effective on the size (p<0.05) in the emulsion technique.

Bu çalışmada, daha önceki araştırmalarımız sonucu elde edilen bilgiler ışığında Bolu Abant İzzet Baysal Üniversitesi Mühendislik Mimarlık Fakültesi Gıda Mühendisliği Bölümü Gıda Mikrobiyolojisi Araştırma Laboratuvarı Kültür Koleksiyonunda bulunan, potansiyel probiyotik özellikleri belirlenmiş ve moleküler genetik yöntemlerle kesin tanısı yapılmış Lactobacillus acidophilus KPb4b suşu kullanılmıştır. Çalışma kapsamında bu potansiyel suşun emülsiyon tekniği, elektrostatik titreşim/damlatma yöntemi ve çift tabaka kaplama (elektrostatik titreşim/damlatma yöntemi ile birinci kaplama üzerine kitosan ile ikinci tabaka kaplama) yöntemi ile mikroenkapsülasyonu gerçekleştirilmiştir. Araştırma sonunda yanıt yüzey yöntemi kullanılarak mikroorganizma canlılığının korunmasında kullanılabilecek uygun parametreler belirlenmeye çalışılmıştır.Araştırma sonuçlarına göre, analiz başlangıç ve sonundaki canlı mikroorganizma sayısı arasındaki fark değerlerinin düşük olması bakterinin analiz koşullarına dayanımının yüksek olduğunu göstermiştir.Araştırma sonuçlarına göre aljinat konsantrasyonunun etkisinin (P<0,01) düzeyinde çok önemli olduğu, ayrıca bu faktörleden aljinat konsantrasyonu ve kitosan konsantrasyonunun interaksiyon etkisinin de (P<0,01) düzeyinde çok önemli olduğu tespit edilmiştir.Araştırma sonuçlarına göre elektrostatik titreşim/damlatma yöntemi ile kaplama ve çift tabaka (Elektrostatik titreşim/damlatma yöntemi + kitosan) ile kaplama yöntemlerinde mikroenkapsül boyutlarına, aljinat konsantrasyonu ve kalsiyum klorür molaritesinin çok önemli düzeyde (P<0,01) etki ettiği sonucuna varılmıştır ve ayrıca emülsiyon tekniğinde aljinat konsantrasyonunun önemli düzeyde (P<0,05) etkili olduğu tespit edilmiştir.
