In this study, it was investigated that the effect of the agmatine, which is a endogenic polyamine formed by the decarboxylation of L-arginine on the increase of the locomotor activity induced by ethanol. In the present study, as a frequently used method in the psycho-pharmalogical studies to evaluate especially the spontaneous behaviours of the test animals occurred by the psychostimulant and sedative effects of the drugs, locomotor Activity (LMA) measurements were employed. Male, adult Swiss-Webster mice (26-36gr.) were used as test animals. It was applied to the mice (in every group, n=8) 0.5 g/kg, 1 g/kg and 2g/kg ethanol intraperitonal for observing the locomotor stimulation. A group of mice were injected with saline. Following the injection, the locomotor activities of the test animals were recorded. After defining the dose of the ethanol (0.5g/kg) that makes a statistically significant locomotor stimulation, agmatine effects were started to investigate. In order to estimate agmatine on the locomotor activity induced by ethanol (0.5g/kg), the test animals were intraperitonally injected by 5 mg/kg, 10 mg/kg and 20mg/kg agmatine (in every group, n=7) 20 minutes before the ethanol application. Immediately after the ethanol injection, the locomotor activities of the test animals were recorded during 30 minutes. The results were compared with saline (n=7) and 0.5g/kg ethanol groups.Agmatine significantly blocked the locomotor activity which was induced by the ethanol (0.5g/kg) within all doses. This is the first data that indicates the blocking effect agmatine on the locomotor activity caused by ethanol. The blocking effect agmatine on the locomotor activity caused by the ethanol may be related to its nitric oxide synthase inhibition or the NMDA receptor blocking properties. In conclusion, our findings are indicating that the agmatine accepted as a new neurotransmitter in the brain is counteracting the pyschostimulan effects of the ethanol and that there is an interaction between the addictive psychostimulant effects of the ethanol and the agmatinergical system.
