The aim of this study was to determine the changes of physical, chemical, microbiological, biochemical and sensory properties of yogurt samples produced by using 8 different inulin and demineralized whey powder (d-whey powder) combinations (inulin % / d-whey powder %; 0% / 0%; 3,0% / 0%; 2,5% / 0,5%; 2,0% / 1,0%; 1,5% / 1,5%; 1,0% / 2,0%; 0,5% / 2,5%; 0% / 3,0%) and a commercial probiotic starter culture (ABT-2) during storage (1., 7., 14., 21. and 28. days) period. According to the findings, effects of the addition of inulin and d-whey powder on the total solid, fat, non-fat solid, ash, protein, viscosity, syneresis, titratable acidity, Lactobacillus acidophilus + Bifidobacterium sp. counts, orotic acid, pyruvic acid, citric acid, lactic acid, acetic acid, butyric acid, hippuric acid amounts, and appearance, consistency and flavor scores were found to be significant at p<0,01 level, while the odor was found significant at p<0,05 level. It was found that the effect of inulin and d-whey powder addition on pH, Streptococcus salivarius subsp. thermophilus, Lactobacillus acidophilus, Bifidobacterium sp. and yeast-mould counts were insignificant statistically.The storage period affected the total solid, fat, non-fat solid, protein, syneresis, S. thermophilus, orotic, pyruvic, citric, lactic and acetic acid amounts, appearance, consistency and flavor scores were significant at the level of p<0,01, while ash, pH, L. acidophilus, Bifidobacterium sp., were significant at the level of p<0,05. On the other hand the effects of storage times viscosity, titratable acidity, L. acidophilus + Bifidobacterium sp., yeast-mould counts, butyric acid, hippuric acid amounts and odor scores were not significant statistically. The analysis of volatile compounds in probiotic yoghurt samples were done using the SPME-GC/MS technique and 53 volatile compounds were determined. It was found that the volatile compounds consist of 2 aldehydes, 6 ketons, 4 alchols, 6 acids, 5 esters, 3 aromatic hydrocarbons, 3 terpens, 12 aliphatic hydrocarbons and 12 other components. It was observed that the live cells of probiotic cultures in all samples contained at the level of 106-107 cfu/g during 28 days of storage. These probiotic yoghurts showed the suitability for consumption with regard to the other parameters. In the sensory evaluations, sample B (3% inulin / 0% d-whey powder) was more acceptable and high scored with respect to meanoverall acceptability (17,30) by panelists.

Bu araştırma, ticari probiyotik starter kültür (ABT-2) ile 8 farklı inülin ve demineralize peyniraltı suyu tozu (d-PAS tozu) (% inülin / % d-PAS tozu ; %0 / %0; %3,0 / %0; %2,5 / %0,5; %2,0 / %1,0; %1,5 / %1,5; %1,0 / %2,0; %0,5 / %2,5; %0 / %3,0) oranı kullanılarak üretilen yoğurtların depolama süresince (1., 7., 14., 21. ve 28. günler) fiziksel, kimyasal, mikrobiyolojik, biyokimyasal ve duyusal özelliklerinde meydana gelen değişmeleri tespit etmek amacıyla yapılmıştır. Elde edilen bulgulara göre, % inülin / % d-PAS tozu ilavesinin kurumadde, yağ, yağsız kurumadde, kül, protein, viskozite, serum ayrılması, titrasyon asitliği, Lactobacillus acidophilus+Bifidobacterium sp. sayısı, orotik asit, pirüvik asit, sitrik asit, laktik asit, asetik asit, bütirik asit ve hippürik asit miktarları ile görünüş kıvam ve lezzet puanları üzerindeki etkisi p<0,01 düzeyinde, koku üzerindeki etkisinin p<0,05 düzeyinde önemli olduğu bulunmuş, pH, Streptococcus salivarius subsp. thermophilus, Lactobacillus acidophilus, Bifidobacterium sp. ve maya-küf sayıları üzerinde ise önemli bir etki meydana getirmediği belirlenmiştir. Depolama periyodunun da kurumadde, yağ, yağsız kurumadde, protein, serum ayrılması, S. thermophilus, orotik, pirüvik, sitrik, laktik ve asetik asit miktarları ile görünüş kıvam ve lezzet puanları üzerinde istatistiksel bakımdan p<0,01 düzeyinde, kül, pH, L. acidophilus ve Bifidobacterium sp. üzerinde p<0,05 düzeyinde önemli olduğu, viskozite, titrasyon asitliği, L. acidophilus+Bifidobacterium sp. ve maya ve küf sayıları ile bütirik asit hippürik asit miktarları ve koku puanları üzerinde ise herhangi bir etki meydana getirmediği tespit edilmiştir. Probiyotik yoğurt örneklerinde uçucu bileşiklerin analizi SPME-GC/MS tekniği kullanılarak yapılmış ve yoğurt örneklerinde 53 uçucu bileşik belirlenmiştir. Uçucu bileşiklerden 2 aldehit, 6 keton, 4 alkol, 6 asit, 5 ester, 3 aromatik hidrokarban, 3 terpen, 12 alifatik hidrokarbon ve 12 diğer bileşenlerden oluşmaktadır. Tüm örneklerin 28 günlük depolama süresince, sağlık üzerinde beklenen yararlı etkinin sağlanabilmesi için gerekli olan 106-107 kob/g düzeyinde canlı mikroorganizma sayısını koruduğu ve diğer parametreler bakımından da tüketime uygunluk gösterdiği tespit edilmiştir. Duyusal değerlendirmeler sonucunda; ortalama en yüksek toplam puanı (17,30) B (%3 inülin / % 0 d-PAS tozu) örneğinin aldığı ve tüm yoğurt örnekleri içerisinde genel kabul edilebilirlik değeri bakımından panelistler tarafından en fazla B örneğinin beğenildiği saptanmıştır.
